The process of Phosphorylation of ADP to ATP coupled with flow of electrons via mitochondrial ETC is termed as oxidative phosphorylation. The electron pairs that originate from oxidation of substrates by NAD -linked dehydrogenases pass through all the three energy conserving sites of Electron transport chain and therefore yield three molecules of ATP. The electron pairs that originate from oxidation of substrates by FAD linked dehydrogenases pass through only two energy conserving sites of ETC and hence yield two molecules of ATP.
Several Mechanisms have been suggested to explain how the energy released during flow of electrons along the ETC is used to phosphorylate ADP to ATP. These mechanisms are i) Chemical Coupling hypothesis
ii) Conformational Coupling Hypothesis iii) Chemi-osmotic Theory.
Chemi-osmotic theory, proposed by Peter Mitchell has been the most accepted theory to explain the mechanism of oxidative phosphorylation.
Complex V in the inner mitochondrial membrane is responsible for oxidative phosphorylation. This complex V is called F1-Fo ATP synthase complex. F1 (head piece) and Fo(base piece), both attached through a stalk. Fo component is not zero but the letter O, which denotes that Fo component of ATP synthase binds oligomycin. Oligomycin inhibits ATP synthase enzyme by blocking the passage of H+ through H+ channel.
