Dr David Sinclair’s lab is collaborating with many other labs working towards the same goal to slow down and even reverse human aging. In this 2021 presentation, Dr Sinclair presents the newest approaches on this. The Main topics are:-

David Sinclair is a professor in the Department of Genetics and co-director of the Paul F. Glenn Center for the Biology of Aging at Harvard Medical School, where he and his colleagues study sirtuins—protein-modifying enzymes that respond to changing NAD+ levels and to caloric restriction—as well as chromatin, energy metabolism, mitochondria, learning and memory, neurodegeneration, cancer, and cellular reprogramming.

Dr Sinclair has suggested that aging is a disease—and that we may soon have the tools to put it into remission—and he has called for greater international attention to the social, economic and political and benefits of a world in which billions of people can live much longer and much healthier lives.

Dr Sinclair is the co-founder of several biotechnology companies (Life Biosciences, Sirtris, Genocea, Cohbar, MetroBiotech, ArcBio, Liberty Biosecurity) and is on the boards of several others.
He is also co-founder and co-chief editor of the journal Aging. He is an inventor on 35 patents and has received more than 35 awards and honors. In 2014, he was on Time Magazine’s list of the “100 Most Influential People in the World,” and listed as Time’s Top 50 in healthcare in 2018.

Dr.Sinclair….are calcium CT scan without contrast……. going to age me …I just learned @72 years young lady… have Mild aortic regurgitation….Soo cardiologist recommends calcium scan….I'm having dental CBCT scans at dentist also ….I'm concerned …. did you have advice … I'm on your protocol and spreading the info….So to calcium scan or not to scan….❤️🙏❤️

  2. Commonsense would tell you that if you can manipulate the mice to age prematurely then it would be logical to think that you could find a way for them to live longer as well. Keep up the good work very much impressed and very hopeful. Keeping you in our prayers. Because I believe that if knowledge of the older generation can be pushed forward into the future that it would give humanity a much better future.

  3. Sinclair got it all wrong. It is the double-strand breaks themselves that drive ageing, and the not the epigenetic changes that histone mobilization for DNA repair cause. SIRTUINS are not anti-ageing proteins. There is absolutely no evidence whatsoever that SIRTUINS exist with the specific purpose of protecting us from the ravages of ageing. SIRTUINS are metabolic stress responders , that adapt the cell for conditions of stress such as starvation and hypoxia. The reason why SIRTUINS increase autophagy, for example, is not because autophay is important for longevity, but because more throrough breakdown of useless cellular components can divert more calorie-rich substances toward energy production. during starvation This is why the primary target of SIRT1 is the PGC1-alpha complex, to increase the generation of mitochondria for more efficient energy production. Likewise, during hypoxia, SIRTUINS are activated to increase SOD(superoxide dismutase), that neutralizes free radicals. This might seem paradoxical, since with less oxygen less fre radicals are created and thus less antioxidants would be needed, but in reality hypoxia generates massive free radicals inside the cell that signal it to committ apoptosis(cell suicide). Now, the enhancement of mitochondrial function and antioxidant defenses might increase halthspan a little bit. But lfespan? Nope. increasing NAD+ in rats with intravenous NR and giving them potent SIRTUIN activators improves their blood glucose and decreases their inflammation, but it does not extend their lives if the rats are fed a healthy diet. It only extends their lives if they are overweight and eating a high-fat diet.

    We know that it is the DNA double-strand breaks that cause ageing and not the epigenetic modification because children who are born with deficient in genes for DNA repair, such ERCC1, GTF2H5, RECQL4, age much faster and have much hiigher rates of cancer than normal children despite having functional SIRTUINS . Deficienvy in the Ku70/80 heterodimer, in particular, responsible for DNA double-strand break repair by non-homologous end joining, causes dramatic acceleration of ageing. They found that DNA double-strand break recognition increases dramatically with species longevity, with mice having the lowest, dogs and cats low but higher than mice, cows and horses intermediate, and humans the most. We also know that bowhead whales, the longest-lived mammals, with a longevity of at least 210 years and possibly 280 years, have extra copies of DNA repair genes, especially ERCC1 compared to humans. The last time I checked, bowhead whales don't take resveratrol and nicotinamide riboside or mononucleotide.

    Increasing NAD+ by different strategies might still not be useless for longevity, NOT because increase NAD+ induces SIRTUINS, but because the PARP enzymes, which recognize DNA damage and mediate the recruitment of multiple DNA repair enzymes, also require NAD+. This is the true benefit of increasing NAD+ not inducing SIRTUINS, which actually inhibit PARP(and vice-versa). Now, this will be of limited benefit, because giving more fuel to your PARPs does not mean they can or will work any harder, but it still might be of some benefit.

    Sinclair is not telling people that his former business, of making SIRTUIN inducers, was sold to a company for a billion Dollars, and the company lost everything when repeated studies after studies showed that there was no life extension from them. some of these inducers were 1,000 X more potent than resveratrol and more orally bioavailable as well, and yet they di nothing in mice, rats and macaques.

    The only two compounds that have repeatedly extended lifespan (and not just healthspan) across all species tested are rapamycin and selegiline and THAT'S IT. Rapamycin is an MTORC-1 inhibitor, and selegiline is a monoamine oxidase B inhibitor. Rapamyxin works by inhibting protein synthesis, which has nothing to do with SIRTUINS. By inhibiting protein synthesis, it decreases cellular division and thus concomitantly DNA damage, while also decreasing protein misfolding. As for selegiline, it works by decreasing the formation of free radicals by blocking the action of monoamine oxidase, whgich generates massive amounts of hydrogen peroxide. It also activates multiple cellular cascades that result in increased intracellular levels of SOD and catalse, mostly by activating the nf-kappaB transcrition factor. Both rapamycin and selegiline are extremely hardcore drugs with a lot of side effects and in the case of selegiline also potentially lethal drug interacitons. Neither are worthy to use as antiageing, unless you have a legitimate medical use for them, such as being a transplant organ receiver or if you have Parkinson's Disease.

    Christ died for our sins according to the Scriptures and that He was buried,and that He rose again the third day according to the Scriptures 1Cor 15 3:8

    Realize that you are a hopeless sinner and trust in Jesus and only Jesus to save you.

    You can say a prayer expressing your gratitude, sorrow for your sins, etc. the words don’t matter, but remember your prayer doesn’t save you.  Your trust in Jesus and what He did for you on the cross is what saves you.
    The moment you trust in Jesus and only Jesus, you are saved.

I wish we can become less disease focused and more treating to reverse or slow aging so disease does not occur or show that these genetic clocks can be reversed or slowed in a human and that available for challenge trials. I feel like we're waiting for a mouse that live for 20 years before we will try anything . Mimicking fasting is not going to do it.

  6. I enjoy listening to your ideas but I feel you often ignore other research and common experience. For example, Dr Rhonda Patrick emphasizes the importance of sauna. Another researcher suggests that many people suffer from a choline (eggs) deficiency. Electrolytes daily especially: Magnesium, Potassium, and Himalayan salt (82 trace elements) add iodine.
    Taurine is especially important for eye health.
    Tyrosine frequently helps people eliminate caffeine (*coffee) or sugar cravings without the highs and crashes associated with caffeine *coffee.
    Most people even meat eaters are often deficient in collagen. Most would benefit from collagen and a high dosage of glycine at night.
    Many people are deficient in L glutamine and would benefit from supplementing plus its very effective at healing an unrecognized leaky gut which may be a root cause of a multitude of problems.
    Most older people are more susceptible to viruses and cancer because their thymus gland diminishes in size but could have benefited by protecting the thymus gland and consuming thymus gland extract. The primary source of natural killer cells is bone marrow and some people could benefit from eating bone broth and bone marrow plus vitamins D, A, and k2mk7, and minerals.
    As men age, they frequently suffer from an enlarged prostate and could benefit from cranberry extract and beta sisterol supplements.
    Most could benefit from a ketogenic diet and intermittent fasting becomes easier with a healthy diet and a proper foundation.
    The RDA for choline is about 3 eggs per day preferably cooked sunnyside up in unrefined virgin coconut oil and add real grass fed butter and himalayan salt.
    Photobiomodulation aka red light therapy is affordable now (*shop around) and extremely effective and beneficial.
    Eliminating or extremely reducing sugar and refined carbohydrates are very beneficial.
    A percussion massager powered by a lithium ion battery is excellent at increasing blood flow to facial muscles and to the scalp.
    Leaning with your head lower than your body like a head stand like a dog down yoga pose can increase blood flow to your brain and scalp.
    Rapid breathing and slow extreme deep breathing for 10 to 20 times can increase oxygenation of the brain. A 10 to 20 cardio work out like proposed by Dr Zach Bush see videos – can improve heart and breathing.
    Daily walks 30 to 60 mins or cardio or lifting weights all are beneficial
    At night: Magnesium, L tryptophan and high dosage glycine and GABA and l glutamine
    In the morning eggs, tyrosine, taurine, and acetyl l carnitine
    Maybe some PQQ and COQ10 UBIQUINOL
