In this interview clip, Dr David Sinclair talks about hot & cold exposure and reveals that sirtuin 3 is induced by cold exposure.
Sirtuins (SIRTs) are nicotinamide adenine dinucleotide (NAD+)-dependent enzymes that regulate energy metabolism, mitochondrial activity and aging. SIRT3, one of the three mitochondrial SIRTs (SIRT3, SIRT4, SIRT5), is localized in the mitochondrial matrix, where it contributes to cellular stress responses . SIRT3 decreases cellular oxidative stress, increases reactive oxygen species (ROS) clearance, enhances the electron transport chain efficacy and stimulates the glutathione production that neutralizes ROS. Overall, SIRT3 modulates the activity of molecules that affect mitochondrial function, metabolism and oxidation responses and protects against the development of age-associated diseases. Calorie restriction, cold exposure and exercise induce SIRT3 expression.
Mitochondrial Sirtuin 3: New emerging biological function and therapeutic target
David Sinclair is a professor in the Department of Genetics and co-director of the Paul F. Glenn Center for the Biology of Aging at Harvard Medical School, where he and his colleagues study sirtuins—protein-modifying enzymes that respond to changing NAD+ levels and to caloric restriction—as well as chromatin, energy metabolism, mitochondria, learning and memory, neurodegeneration, cancer, and cellular reprogramming.
Dr David Sinclair has suggested that aging is a disease—and that we may soon have the tools to put it into remission—and he has called for greater international attention to the social, economic and political and benefits of a world in which billions of people can live much longer and much healthier lives.
Dr David Sinclair is the co-founder of several biotechnology companies (Life Biosciences, Sirtris, Genocea, Cohbar, MetroBiotech, ArcBio, Liberty Biosecurity) and is on the boards of several others.
He is also co-founder and co-chief editor of the journal Aging. He is an inventor on 35 patents and has received more than 35 awards and honors. In 2014, he was on Time Magazine’s list of the “100 Most Influential People in the World,” and listed as Time’s Top 50 in healthcare in 2018.

ok. so, I have an autoimmune disease. A couple, actually, but one of my portrayals of underlying autoimmune issues is my psoriasis. My psoriasis was well treated through poison…. the doctor I had decades ago said it was overactive T cells, and that by poisoning the body it gives the T cells somethng to do. After the T cells have done what they think they need to do, they will settle down.
So, due to that theory, I had two prescription topical poisons as well as prescribed UV therapy. It did work. A decade later, I'm not using the topical poisons anymore because it was under control, but then, for reasons unknown to me, the psoriasis started raging again, covering at least 30% of my body. Previous treatment didn't work. Unintentionally, I got a SEVERE sunburn. I mean SEVERE, all over my body. I had gone to a weekend long family reunion at a lake…. Psoriasis went away. Just like that!
Also, as another poison therapy, there is some research on hookworms. They are bad when there's a lot. They're good when there's a few. Do you sell hookworms? Just kidding. But, going by the poison theory, this cold therapy might be related. The poison theory would be that if you give the body something to unrelated to fight then the auto immune disease goes away? Thoughts?
